高效液相色谱已广泛应用于许多领域,但由于填料昂贵,大都作为分析的手段,而极少用于制备分离。我们将青岛海洋化工厂生产的薄层色谱用硅胶H作为制备型液相色谱柱填料进行了测试,发现其具有良好的性能,并用三种不同柱径的色谱柱观察了柱径对柱效的影响。实验部分仪器和试剂: 仪器:自制PC-80型制备液相色谱仪,紫外检测器(254nm),流动池体积15μl,柱出口和流动池之间体积约118μl。填料:青岛海洋化工厂薄层色谱用硅胶H,粒径10—40μm,含水量6%。另按一般常规法处理制成含水量分别为10%与20%的硅胶H。
High performance liquid chromatography has been widely used in many areas, but due to the expensive packing, mostly as a means of analysis, and rarely used for the preparation of separation.
