本文从人食管癌组织提取高分子量DNA,在体外对大鼠成纤维细胞Rat—1进行转染,建立了人食管癌转化细胞系。经软琼脂培养,分子杂交和裸鼠致瘤性等实验证明,这些转化细胞系具有恶性表型,具有肿瘤形成能力,存在人的DNA序列。本细胞系将为深入研究人食管癌癌基因奠定了基础。
In this paper, high molecular weight DNA was extracted from human esophageal cancer tissue and rat fibroblast Rat-1 was transfected in vitro to establish a human esophageal cancer cell line. Experiments on soft agar, molecular hybridization and tumorigenicity in nude mice have demonstrated that these transformed cell lines have a malignant phenotype, have the ability to form tumors, and have human DNA sequences. The cell line will lay the foundation for further study of human esophageal oncogene.
