目的:检验我们设计的DNA梯度化分析软件在细胞凋亡定量检测中的应用价值 方法:用Camptothesin(CAM)诱导U937细胞株凋亡,通过该系统计算DNA琼脂糖凝胶电泳后DNA的梯度值,并与凋亡细胞胞浆寡聚核小体的Elisa检测值进行相关性分析.结果:随着CAM浓度的增加,DNA琼脂糖凝胶电泳后梯度化程度越明显,测出的梯度值也越大,而且与胞浆寡聚核小体的Elisa检测值呈正相关(r=0.989,p<0.01)结论:利用DNA梯度化分析系统可对琼脂糖凝胶电泳后的凋亡细胞DNA片段,进行定量分析,提供简便直接的方法.
OBJECTIVE: To test the value of DNA gradient analysis software designed by us in the quantitative detection of apoptosis. Methods: The apoptosis of U937 cells was induced by Camptothesin (CAM), and the gradient of DNA after DNA agarose gel electrophoresis , And correlated with the Elisa values of apoptotic cytoplasmic oligomer nucleosomes.Results: The gradient of DNA agarose gel electrophoresis was more obvious with the increase of CAM concentration, and the gradient value (R = 0.989, p <0.01) .Conclusion: A DNA gradient analysis system can be used to detect DNA fragments of apoptotic cells after agarose gel electrophoresis, Quantitative analysis provides an easy and straightforward way.
