G蛋白偶联受体激酶相互作用蛋白1/2[G-protein-coupled receptor(GPCR)kinase interacting protein 1/2,GIT1/2]属于多结构域蛋白质。它们参与多种细胞生物学过程:(1)通过其ARF GTPase-激活蛋白(ARF GTPase-activating protein,ARF-GAP)活性调节细胞骨架动态变化;(2)参与G蛋白偶联受体的内化及细胞膜转运;(3)作为支架蛋白调控信号转导。GIT蛋白家族在骨代谢、血管稳态、神经发育以及肿瘤等生理或病理条件下发挥关键的调控作用。本文着重就近年来GIT蛋白家族在生理与病理方面的调控作用作一综述。
G protein-coupled receptor (GPCR) kinase interacting protein 1/2, GIT1 / 2 belongs to the multidomain protein. They are involved in a variety of cellular biological processes: (1) regulation of cytoskeletal dynamics through their ARF GTPase-activating protein (ARF-GAP) activity; (2) internalization of G protein-coupled receptors And cell membrane transport; (3) as a scaffold protein regulatory signal transduction. The GIT protein family plays a key regulatory role in bone metabolism, vascular homeostasis, neurodevelopment, and tumor or other physiological or pathological conditions. This article focuses on the GIT protein family in recent years, the physiological and pathological aspects of the regulatory role are reviewed.
