本文报导以粒花冠小月螺(Lunella coronata granulata Gmelin)为材料,粗提其消化酶液,用以对海藻细胞的解壁作用和原生质体的制备进行了研究。结果表明,此螺消化酶液对海藻细胞的解壁有较好的效果,能成功地制备大量分离细胞和原生质体,成活率达80—90%。文中还对该消化酶液的性质以及影响藻细胞解壁作用的若干条件(例如,浓度、pH值、酶解温度、酶解液的渗透势以及低温贮藏等)作了论述。为该消化酶液在紫菜酶法育苗,科学研究、生物工程技术以及教学方而,提供参考资料。
In this paper, Lunella coronata granulata Gmelin was used as a material to crudely digest its digestive enzyme solution to study the decontamination of seaweed cells and the preparation of protoplasts. The results showed that this solution of digestive enzymes had a good effect on the solution of seaweed cells, and successfully prepared a large number of isolated cells and protoplasts with a survival rate of 80-90%. The nature of the digestive enzyme solution, as well as the conditions affecting the disaggregation of algae cells (eg, concentration, pH value, enzymatic hydrolysis temperature, osmolality of the hydrolyzate, and cryogenic storage, etc.) are also discussed. For the digestive enzymes in the laver enzymatic nursery, scientific research, bio-engineering technology and teaching, while providing reference materials.
