目的:研究蒲黄提取物对氧化低密度脂蛋白(Ox-LDL)诱导人脐静脉内皮细胞(HUVECs)脂质过氧化的影向。方法:体外培养人脐静脉内皮细胞,采用MTT法测定不同浓度蒲黄提取物对内皮细胞有无毒性。根据MTT法的结果,采用蒲黄提取物(10mg/L,50mg/L,100mg/L)预处理细胞24h,再加入100mg/L Ox-LDL造成细胞损伤,检测细胞培养液中乳酸脱氢酶(LDH)的活性、丙二醛(MDA)的含量及超氧化物歧化酶(SOD)的活力。结果:Ox-LDL损伤组LDH的活性、MDA的含量显著升高,SOD的活力显著降低,与正常对照组比较有显著差异(P<0.05);10、50、100mg/L蒲黄提取物可显著降低LDH的活性、MDA的含量,提高SOD的活力。结论:蒲黄提取物可通过抗脂质过氧化,清除自由基起到保护血管内皮细胞的作用。
Objective: To study the effect of Puhuang extract on lipid peroxidation of human umbilical vein endothelial cells (HUVECs) induced by Ox-LDL. Methods: Human umbilical vein endothelial cells were cultured in vitro. The toxicity of different concentrations of Puhuang extract to endothelial cells was determined by MTT assay. According to the results of MTT method, the cells were pretreated with Puhuang extract (10mg / L, 50mg / L, 100mg / L) for 24 h and then added with 100 mg / L Ox-LDL to induce cell injury. Lactate dehydrogenase (LDH) activity, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity. Results: The activity of LDH, the content of MDA and the activity of SOD in Ox-LDL injury group were significantly lower than those in normal control group (P <0.05). The extract of Puhuang (10,50,100 mg / L) Significantly reduce the activity of LDH, MDA content, improve the vitality of SOD. Conclusion: Puhuang extract can protect vascular endothelial cells by anti-lipid peroxidation and free radical scavenging.
