目的建立小柴胡汤正丁醇有效部位整体特征的质量评价方法。方法采用高效液相蒸发光散射检测器,Diamonsil C18(2)(250mm×4.6mm,5μm)色谱柱,以乙腈-体积分数为0.05%的甲酸水溶液为流动相进行梯度洗脱,建立小柴胡汤和各药材正丁醇有效部位指纹图谱。结果经评价软件计算相似度大于0.9,对21个共有峰进行了药材归属,并指认了其中的6个色谱峰。结论建立的小柴胡汤正丁醇有效部位的HPLC-ELSD指纹图谱准确稳定、重复性好,且能较好地判别有效部位主要色谱峰与药材化学组分间的相关性,为阐明有效部位的化学物质基础提供科学依据。
OBJECTIVE To establish a method for quality assessment of the overall features of the effective fraction of n-butanol in Xiao Chai Hu Tang. Methods A gradient elution was performed on a Diamonsil C18 (2) (250 mm × 4.6 mm, 5 μm) column with a mobile phase of acetonitrile-0.05% And the medicinal n-butanol effective part of the fingerprint. Results The calculated software similarity was greater than 0.9. The 21 common peaks were assigned to medicinal herbs and six of them were identified. Conclusion The established HPLC-ELSD fingerprints of n-butanol in Xiaochaihu Tang is accurate and reproducible, and it can well discriminate the correlation between the main chromatographic peak of effective part and the chemical components of medicinal materials. In order to elucidate the chemical constituents of effective parts, Basis to provide a scientific basis.
