目的建立获得高纯度HSC/HPC细胞群的方法,用于研究造血干/祖细胞(HSC/HPC)的功能与生物学特性。方法利用免疫磁珠分选法(MACS)从新鲜脐血和骨髓穿刺液中富集CD34+细胞,再用荧光激活细胞分选术(FACS)分选纯化CD34+/CD38-和CD34+/CD38+细胞亚群。结果MACS分选的CD34+细胞群纯度可达95%;FACS分选的CD34+/CD38-和CD34+/CD38+两细胞亚群纯度均可达99%以上。结论联合使用MACS和FACS可在数小时内大量纯化HSC/HPC及其中重要细胞亚群。
Objective To establish a method for obtaining high-purity HSC/HPC cell population for studying the functional and biological characteristics of hematopoietic stem/progenitor cells (HSC/HPC). METHODS: CD34+ cells were enriched from fresh umbilical cord blood and bone marrow aspirate by immunomagnetic bead sorting (MACS), and then CD34+/CD38- and CD34+/CD38+ subpopulations were sorted by fluorescence-activated cell sorting (FACS). . Results The purity of CD34+ cell population with MACS sorting was up to 95%. The purity of CD34+/CD38- and CD34+/CD38+ subpopulations with FACS sorting was up to 99%. Conclusion The combination of MACS and FACS can be used to massively purify HSC/HPC and its important cell subpopulations within a few hours.
