目的 :建立反相高效液相色谱法测定甘草甜素粉剂和四君子汤灌胃大鼠血浆中的甘草酸铵。方法 :以联苯为内标 ,甲醇 -水 -醋酸 (80∶19∶1)为流动相 ,采用ODS - 3色谱柱 (15 0mm× 4 6mm ,5 μm) ,检测波长 :2 5 0nm ,流速 :1 0mL·min-1。结果 :最低检测限为 2ng ,最低检测浓度为 0 2 2 8μg·mL-1,日间和日内RSD <10 % ,回收率为 92 46 %~10 3 2 % ,RSD为 3 8%~ 6 0 % ,在 1 2 2 4~ 38 6 μg·mL-1和 40 8~ 146 8 8μg·mL-1范围内均呈良好的线性关系。结论 :本法简便、快速、灵敏而准确 ,适用于大鼠灌胃不同甘草酸铵制剂后血浆中甘草酸铵的测定 ,为“复方效应成分动力学”研究奠定了方法学基础。
Objective: To establish an RP-HPLC method for determination of glycyrrhizic acid in rat plasma by glycyrrhizin powder and Sijunzi decoction. Methods: Biphenyl was used as the internal standard and the mobile phase was methanol - water - acetic acid (80:19:1). The mobile phase was ODS - 3 (150 mm × 4 6 mm, 5 μm) : 10 mL · min-1. Results: The lowest detection limit was 2 ng, the lowest detection limit was 0 2 2 8 μg · mL-1. The RSD was less than 10% in daytime and day, the recovery was 92 46% -103 2%, RSD was 38% -6 0 %, With a good linear relationship in the range of 1 2 2 4 ~ 38 6 μg · mL -1 and 40 8 ~ 146 8 8 μg · mL -1. Conclusion: The method is simple, rapid, sensitive and accurate and is suitable for the determination of ammonium glycyrrhizinate in rats after intragastric administration of different formulations of glycyrrhizin, which lays the foundation of methodology for the study of “compound effect component kinetics”.
