ras基因家族是癌基因家族中较为活跃的一支,近年来已有许多实验证明,在多种肿瘤中都能检测到ras基因的异常激活和表达。本实验分别构建了含不同ras基因片段的重组质粒pN-ras 8.8、pEJ-Ha-ras 6.6和pV-kiras 1.0,并从中提纯探计进行放射性标记,用斑点印迹杂交和Northern印迹杂交检测了人类大肠癌细胞系(Lovo)中ras基因的表达情况,以期为探索人类大肠癌发生和发展的机理提供参考。
The ras gene family is one of the more active oncogene families. In recent years, many experiments have proved that the ras gene can be abnormally activated and expressed in many tumors. In this study, recombinant plasmids pN-ras 8.8, pEJ-Ha-ras 6.6 and pV-kiras 1.0 containing different ras gene fragments were constructed respectively and radioactively labeled from the purified ragger probes. Human and human samples were detected by dot blot hybridization and Northern blot hybridization To investigate the expression of ras gene in human colorectal cancer cell line Lovo in order to provide reference for exploring the mechanism of human colorectal cancer occurrence and development.
