目的 了解姜黄素与羟基脲合用对人白血病 K5 6 2细胞的体外作用。 方法 采用 MTT法测定药物的体外杀伤作用 ,应用 AO/EB荧光染料染色观察药物诱导细胞凋亡 ,金氏公式进行联合用药分析。 结果 姜黄素 5 ,10μg/ml,羟基脲 2 5 ,5 0μg/m l同时给药 ,对 K5 6 2细胞可产生单纯相加的协同杀伤效果。姜黄素与羟基脲同时给药诱导 K5 6 2细胞凋亡率高于羟基脲单独用药 ,低于姜黄素单独用药。 结论 姜黄素与羟基脲同时联合用药可产生单纯相加的协同杀伤效用 ,姜黄素可部分逆转 K5 6 2细胞对羟基脲诱导凋亡的抗药性。
Objective To understand the in vitro effects of curcumin combined with hydroxyurea on human leukemia K562 cells. Methods The in vitro killing effect of the drug was determined by MTT method. Drug-induced apoptosis was observed by AO/EB fluorescent dyeing. Kim’s formula was used for combined drug analysis. RESULTS Curcumin 5, 10 μg/ml, hydroxyurea 25, 50 μg/ml were administered at the same time, and a synergistic killing effect on K562 cells could be achieved. The apoptosis rate of K562 cells induced by simultaneous administration of curcumin and hydroxyurea was higher than that of hydroxyurea alone, lower than that of curcumin alone. Conclusion The combined use of curcumin and hydroxyurea can produce a synergistic effect with only additive effects. Curcumin can partially reverse the resistance of K562 cells to hydroxyurea-induced apoptosis.
