目的:研究TNF-α诱导人蜕膜细胞异常凋亡后基因表达谱的变化,为了解病理妊娠蜕膜细胞基因表达及药物治疗研究提供基础。方法:体外常规进行人蜕膜细胞培养,选用适宜浓度的TNF-α诱导建立人蜕膜细胞异常凋亡模型,采用人全基因组17Kc DNA基因芯片分析TNF-α作用后的基因表达谱变化情况,分析基因的变化趋势。结果:TNF-α诱导蜕膜细胞后检出基因9 320条,上调基因36条,下调基因48条。表达上调的已知基因主要为细胞周期依赖性蛋白激酶和抑制剂基因、DNA损伤及修复基因,另外还有氧化压力和炎症损伤基因等。表达下调的基因与细胞营养、细胞生长、细胞周期及信号转导相关。结论:TNF-α诱导蜕膜细胞异常凋亡。
OBJECTIVE: To study the changes of gene expression profile induced by TNF-α induced apoptosis in human decidual cells, and to provide a basis for understanding the gene expression of decidual cells and drug therapy in pathological pregnancy. Methods: Human decidual cells were cultured in vitro. Abnormal apoptosis models of human decidual cells were induced by appropriate concentration of TNF-α. The changes of gene expression profile after TNF-α treatment were analyzed by human whole genome 17Kc DNA microarray. Analyze the changing trend of genes. Results: After the decidual cells were induced by TNF-α, 9 320 genes were detected, 36 genes were up-regulated and 48 genes were down-regulated. Known genes whose expression is up-regulated are mainly cell cycle-dependent protein kinase and inhibitor genes, DNA damage and repair genes, in addition to oxidative stress and inflammatory injury genes. Down-regulated genes are associated with cell nutrition, cell growth, cell cycle and signal transduction. Conclusion: TNF-α induces abnormal apoptosis of decidual cells.
