检测线粒体膜通透性转运孔(MPT)的开放抑制剂环孢菌素A(CSA)对芹菜夜蛾核型多角体病毒(Syngrapha falcifera multiplenuclear polyhedrosis virus,SfaMNPV)诱导诱导斜纹夜蛾(Spodoptera litura Cells,SL-1)细胞凋亡的影响。用不同浓度CSA和SfaMNPV单独或联合处理SL-1细胞,通过细胞形态学观察、DNA凝胶电泳法、PI染色流式细胞仪鉴定细胞凋亡,应用罗丹明123(Rh123)染色流式细胞仪测定线粒体跨膜电位(ΔΨm)。CSA明显抑制了SfaMNPV诱导的SL-1细胞凋亡,CSA还抑制了SfaMNPV诱导的SL-1细胞线粒体膜电位(ΔΨm)下降。CSA抑制了SfaMNPV诱导SL-1细胞的ΔΨm下降,可能是亲环素-D(CyP-D)与CSA的结合导致MPT的关闭,从而抑制细胞凋亡。
Cytosporin A (CSA), an open inhibitor of mitochondrial membrane permeability transition pore (MPT), was used to induce the apoptosis of Spodoptera litura Cells induced by Synechocystis multiple nuclear polyhedrosis virus (SfaMNPV) , SL-1) apoptosis. SL-1 cells were treated with different concentrations of CSA and SfaMNPV alone or in combination, and cell apoptosis was identified by morphological observation, DNA gel electrophoresis and flow cytometry with PI staining. The cells were stained with Rh123 staining flow cytometry Mitochondrial transmembrane potential (ΔΨm) was measured. CSA significantly inhibited SfaMNPV-induced SL-1 cell apoptosis, and CSA also inhibited SfaMNPV-induced decrease in mitochondrial membrane potential (ΔΨm) of SL-1 cells. CSA inhibited SfaMNPV-induced decrease in ΔΨm in SL-1 cells. It is likely that the binding of cyclophilin-D (CyP-D) to CSA resulted in the closure of MPT, thereby inhibiting apoptosis.
