研究Kv 1.4和Kv 4.3蛋白在狗心室外向钾电流形成中的作用。利用免疫印迹和免疫组化技术 ,证实了在狗心室肌细胞均有Kv 1.4和Kv 4.3蛋白的表达。电生理实验也提示在狗Ito电流中有Kv 1.4和Kv 4.3的成分。与猫、鼠心脏 (Kv1.4表达仅限于心室内膜 )不同 ,狗的Kv 1.4在整个心室肌层均有表达 ,故对Ito通道的功能有更重要的作用。在狗的Ito通道中 ,β亚单位起着主要的作用 ,可用来解释在狗心肌上所表现的“表型 -转换”现象。
To investigate the role of Kv1.4 and Kv4.3 proteins in the outward potassium current in dogs. Immunoblotting and immunohistochemistry confirmed the expression of Kv 1.4 and Kv 4.3 proteins in dog ventricular myocytes. Electrophysiological experiments also suggest that there is a composition of Kv 1.4 and Kv 4.3 in dog Ito current. Unlike the cat and rat hearts (Kv1.4 expression is restricted to the ventricular endocardium), Kv1.4 in dogs is expressed throughout the myenteric myocardium and therefore plays a more important role in Ito channel function. In the dog’s Ito channel, the beta subunit plays a major role in explaining the “phenotype-switching” phenomenon that occurs in the dog’s myocardium.
