目的探讨ANXA2、VEGF在HepG2肝癌细胞系中的表达及意义。方法应用Transwell(细胞体外侵袭实验)、WesternBlot检测在不同5-Fu剂量干扰下,HepG2肝癌细胞系中ANXA2、VEGF的表达及细胞侵袭力的变化。结果 Transwell检测HepG2肝癌细胞随着5-Fu剂量的增加侵袭力明显降低,各剂量组间差异具有统计学意义(P<0.05);WesternBlot方法检测HepG2肝癌细胞中ANXA2、VEGF蛋白质的表达随着5-Fu剂量的增加逐渐下降,各剂量组间差异具有显著统计学意义(P<0.01),且两者具有相关性r=0.856。结论 ANXA2、VEGF在HepG2肝癌细胞系的侵袭转移机制中可能具有协同促进作用。
Objective To investigate the expression and significance of ANXA2 and VEGF in HepG2 hepatocellular carcinoma cell lines. Methods Transwell (cell invasion assay) and Western Blot were used to detect the expression of ANXA2 and VEGF and the invasiveness of HepG2 cells in HepG2 cells with different doses of 5-Fu. Results The invasiveness of HepG2 hepatocarcinoma cells was significantly decreased with the increase of 5-Fu dosage by Transwell assay (P <0.05). The protein expression of ANXA2 and VEGF in HepG2 hepatocarcinoma cells was detected by Western Blot with 5 -Fu dosage decreased gradually, the difference between each dose group was statistically significant (P <0.01), and the correlation between the two was r = 0.856. Conclusion ANXA2 and VEGF may play synergistic roles in the invasion and metastasis of HepG2 hepatocellular carcinoma cell lines.
