目的建立国境口岸重要蚊类分子鉴定方法。方法针对口岸10种重要蚊类,包括淡色库蚊(Culex pipiens pallens)、致倦库蚊(Culex pipiens quinquefasciatus)、凶小库蚊(Culex modestus)、三带喙库蚊(Culex tritaeniorhynchus)、埃及伊蚊(Aedes aegypti)、白纹伊蚊(Aedes albopictus)、背点伊蚊(Aedes dorsalis)、刺扰伊蚊(Aedes vexans)、中华按蚊(Anopheles sinensis)和骚扰阿蚊(Armigeres subalbatus)设计特异性引物或通用引物进行核酸扩增,测序后与Gen Bank中的序列比对分析。结果测定的10种重要蚊类均确认为相应蚊种;利用CO I基因的系统发育分析的结果显示与传统的形态鉴定结果一致。结论分子鉴定能准确进行蚊类鉴定,CO I基因在蚊类鉴定中可以作为有效的分子遗传标记。
Objective To establish molecular identification methods of important mosquitoes at border crossings. Methods A total of 10 important mosquitoes at the port, including Culex pipiens pallens, Culex pipiens quinquefasciatus, Culex modestus, Culex tritaeniorhynchus, Aedes aegypti, Aedes albopictus, Aedes dorsalis, Aedes vexans, Anopheles sinensis and Armigeres subalbatus Nucleic acid amplification using a primer or a universal primer, and comparing the sequence with GenBank after sequencing. The results of the determination of the 10 kinds of important mosquitoes were identified as the corresponding mosquito species; the use of COI gene phylogenetic analysis results show that with the traditional morphological identification results. Conclusion Molecular identification can accurately identify mosquitoes. CO I gene can be used as an effective molecular genetic marker in the identification of mosquitoes.
