目的:快速鉴定光叉叶委陵菜Potentilla bifurca var.glabrata中的酪氨酸酶抑制成分。方法:利用活性-高效液相色谱跟踪方法(bioassay-linked HPLC method),通过接收器接样,并用酪氨酸酶抑制强度,快速查找有效成分。结果:光叉叶委陵菜的甲醇提取物及其正丁醇萃取物显示较强的酪氨酸酶抑制作用。从光叉叶委陵菜的正丁醇萃取物中分离并鉴定一种黄酮类化合物,即槲皮素-4′-O-β-D-葡萄糖苷。与已知的酪氨酸酶抑制剂熊果苷(arbutin,IC50=0.28mmol.L-1)相比,槲皮素-4′-O-β-D-葡萄糖苷化合物具有更强的酪氨酸酶抑制作用。其IC50值为0.0019mmol.L-1。结论:该方法对中药及其复方药快速确定有效成分提供分析手段。
OBJECTIVE: To rapidly identify the tyrosinase-inhibitory components of Potentilla bifurca var.glabrata in the leaves of Potentilla. Methods: The active-high-performance liquid chromatography method (bioassay-linked HPLC method) was used to accept the sample through the receiver and use tyrosinase to inhibit the strength to quickly find the active ingredient. RESULTS: The methanol extract and its n-butanol extract of P. oleifera showed strong tyrosinase inhibition. A flavonoid compound, quercetin-4’-O-β-D-glucoside, was isolated and identified from the n-butanol extract of Potentilla viridis. Compared to known tyrosinase inhibitor arbutin (IC50 = 0.28 mmol.L-1), quercetin-4’-O-β-D-glucoside compound has stronger tyrosine Acidase inhibition. Its IC50 value is 0.0019 mmol.L-1. Conclusion: This method provides an analytical tool for the rapid determination of effective components of traditional Chinese medicine and its compound medicine.
