目的 探讨在大鼠脑发育期补充必需脂肪酸兴奋性氨基酸 (EFA)对脑组织中EAA含量的影响。方法 断乳大鼠 (1个月 ) 3 0只 ,雌雄不拘 ,分为正常组、EFA缺乏组和补充鱼油组 ,分别以不同饲料喂养 3个月 ,取出新鲜的大脑皮层和海马组织 ,测定天门冬氨酸、谷氨酸的含量 ;尼氏染色和嗜银染色观察大脑皮质和海马细胞形态结构的变化。结果 与正常组相比 ,EFA缺乏组的EAA含量均显著升高 (P <0 0 5 ) ,补充鱼油组无明显差异 ;与补充组相比 ,EFA缺乏组大脑皮质大锥体细胞数目明显减少、排列不规则、稀疏、轴突纤维短 ,海马细胞层数较少、体积也较小。结论 发育期EFA缺乏会导致脑组织中EAA含量明显升高 ,神经形态异常。
Objective To investigate the effect of dietary fatty acid excitatory amino acid (EFA) supplementation on brain EAA content during rat brain development. Methods 30 weaned rats (1 month) were randomly divided into normal group, EFA deficient group and fish oil supplemented group. The rats were fed with different diets for 3 months, fresh cortex and hippocampus were removed, Aspartic acid and glutamic acid. The morphological changes of cerebral cortex and hippocampus were observed by Nissl staining and argyrophilic staining. Results EAA levels in EFA deficient group were significantly higher than those in normal control group (P <0.05), and there was no significant difference in fish oil supplemented with EAF group. Compared with the supplemented group, the number of large pyramidal neurons in cerebral cortex significantly decreased in EFA deficient group , Arranged irregular, sparse, axon fibers short, the number of layers of hippocampus cells less, the volume is smaller. Conclusion The lack of EFA during development leads to the marked increase of EAA content in brain tissue and abnormal neuromorphology.
