目的 :检测脑胶质瘤细胞体外药敏及P -糖蛋白的表达 ,为临床选择化疗药物制定方案提供依据。方法 :采用MTT法检测 30例脑胶质瘤患者肿瘤细胞 ,对化疗药物的敏感性以及采用免疫组化方法检测脑胶质瘤细胞的P -糖蛋白的表达。结果 :30例脑胶质瘤患者肿瘤细胞体外药敏显示 :高三尖杉酯碱 (H)、长春新碱(VCR)、阿糖胞苷 (Ara -C)、足叶乙甙 (Vp - 16 )、柔红霉素 (DNR)、米托葸昆 (MX)、平阳霉素 (SP)、阿克拉霉素 (ACR)、阿霉素 (ADM )、环已亚硝脲 (CCNU)等化疗药物对肿瘤细胞均有一定的抑制作用 ,以CCNU、ADM、ACR、DNR、MX、Vp - 16抑制作用较强。检测 2 8例患者脑胶质瘤细胞P -糖蛋白表达 ,2 4例表达(- ) ,4例表达 (+) ,其中 3例为复发患者。结论 :胶质瘤细胞体外药敏及P -糖蛋白表达的检测 ,可为临床指导用药 ,制定化疗方案 ,提供重要参考实用价值。
OBJECTIVE: To detect the drug sensitivity and the expression of P - glycoprotein in glioma cells in vitro so as to provide evidence for the clinical selection of chemotherapeutics. Methods: Tumor cells from 30 patients with glioma were assayed by MTT assay. The sensitivity to chemotherapy drugs and the expression of P - glycoprotein in glioma cells were detected by immunohistochemistry. Results: The in vitro susceptibility of tumor cells in 30 patients with gliomas showed that Hp, VCR, Ara - C, Vp - 16 (DNR), mitoxantrone (MX), pingyangmycin (SP), aclacinomycin (ACR), doxorubicin (ADM) The drug has a certain inhibitory effect on tumor cells, with CCNU, ADM, ACR, DNR, MX, Vp - 16 inhibition stronger. The expression of P - glycoprotein in 24 glioma cells was detected in 24 cases (-) and in 4 cases (+), of which 3 cases were recurrent. Conclusion: The detection of glioma cells in vitro and the detection of P - glycoprotein may provide an important reference value for clinical guidance and development of chemotherapy regimens.
