目的研究广藿香醇对东莨菪碱所致获得性学习记忆障碍小鼠学习记忆功能的影响。方法采用Morris水迷宫检测小鼠学习记忆能力,广藿香醇(80、40、20mg/kg)连续预防给药10d,训练前15minip给予东莨菪碱(1.5mg/kg)。行为学测试完成后,脑匀浆检测乙酰胆碱脂酶(AchE),丁酰胆碱脂酶(BuchE)以及胆碱乙酰转移酶(Chat)活性,免疫组化检测脑M1受体水平。结果与模型组相比,广藿香醇(80、40mg/kg)组小鼠学习记忆能力显著增强,脑AchE活性显著下降,而Chat活性显著提高,脑内M1受体水平也显著提高。结论广藿香醇具有促进东莨菪碱小鼠学习记忆的作用,该作用可能与其抑制脑内AchE活性,提高Chat活性和M1受体水平有关。
Objective To study the effects of patchoulol on learning and memory in mice with acquired learning and memory impairment induced by scopolamine. Methods Morris water maze was used to detect the learning and memory ability of mice. Patchouli alcohol (80, 40, 20 mg / kg) was given for 10 days and scopolamine (1.5 mg / kg) for 15 minutes before training. After behavioral testing, brain homogenate was used to detect the activity of acetylcholinesterase (AchE), butyrylcholinesterase (BuchE) and choline acetyltransferase (Chat). The levels of brain M1 receptor were detected by immunohistochemistry. Results Compared with the model group, the learning and memory abilities of patchouli alcohol group (80,40 mg / kg) were significantly increased, the activity of AchE decreased significantly, and the activity of Chat increased significantly. The level of M1 receptor in brain increased significantly. Conclusions Patchouli alcohol can promote the learning and memory of scopolamine in mice, which may be related to the inhibition of AchE activity in the brain, the increase of the activity of Chat and the level of M1 receptor.
