本文涉及组织培养领域的一种新技术。它的基本特征是试管苗或脱毒苗生长在特制的敞口容器中。培养基和外植体由一层粉末材料覆盖,它可以有效地阻止微生物污染而不阻止脱毒苗茎叶的生长和钻出粉末层,阻止水分外渗而不阻止氧气渗入。由于茎叶暴露于容器外部,接受自然光的直射和干燥通风等条件的锻炼,试管苗生长矮壮、色绿,不用练苗而移栽成活率很高,其素质接近常规繁殖的幼苗。暴露培养法可以克服传统组培技术由于封闭系统方式而造成的幼苗娇弱,移栽后成活率低以及成本过高等固有缺点。
This article deals with a new technique in the field of tissue culture. Its basic feature is the growth of test-tube seedlings or virus-free seedlings in specially prepared open containers. The medium and explants are covered with a layer of powdered material that can effectively prevent microbial contamination without inhibiting the growth of stems and leaves of the virus-detoxified seedlings and drilling out of the powder layer, preventing extravasation of moisture without impeding oxygen infiltration. As stems and leaves exposed to the outside of the container to accept the direct sunlight and dry ventilation and other conditions of exercise, test tube seedlings grow strong, green, without seedlings and transplant survival rate is very high, its quality close to conventional breeding seedlings. The exposure culture method can overcome the inherent disadvantages of traditional tissue culture techniques such as delicate seedlings, low survival rate after transplanting and high cost due to the closed system.
