目的 :探讨蛋白激酶C(PKC)何种亚型对鼻息肉组织中嗜酸性粒细胞 (EOS)浸润增多具有调控作用。方法 :采用原位杂交法和免疫组织化学MGG染色 ,观察 2 6例鼻息肉组织中PKC几种亚型PKCα、PKCβ1、PKCβ2 、PKCγ的表达 ,分析何种亚型在鼻息肉EOS增多中起调控作用。 结果 :2 6例鼻息肉组织EOS中均有PKC表达 ,且与Bcl 2mRNA及其蛋白成正相关 (r1=0 .0 87 5 ,r2 =0 .0 82 3,P <0 .0 1) ,其中PKCα为强表达 ,而PKCβ1、PKCβ2 为弱表达 ,PKCγ则不表达。结论 :EOS增多主要是由于激活了PKC这一信号传导途径 ,使EOS凋亡受到抑制、生存延长 ,致使其数量增多 ,且主要是PKC亚型中的PKCα在鼻息肉组织EOS增多中起调控作用
Objective: To investigate which subtype of protein kinase C (PKC) plays a role in the regulation of eosinophil infiltration in nasal polyps. Methods: In situ hybridization and immunohistochemical staining of MGG were used to observe the expression of PKCα, PKCβ1, PKCβ2 and PKCγ in 26 cases of nasal polyps, and to analyze which subtype plays a role in the increase of EOS in nasal polyps effect. Results: The expression of PKC in EOS of 26 cases of nasal polyps was positively correlated with the expression of Bcl-2 mRNA and protein (r1 = 0.087 5, r2 = 0.082 3, P <0.01) PKCα is strongly expressed, whereas PKCβ1 and PKCβ2 are weakly expressed, while PKCγ is not expressed. CONCLUSIONS: The increase of EOS is mainly attributed to activation of PKC signaling pathway, which inhibits the apoptosis of EOS and prolongs its survival, resulting in an increase in the number of PKCα in PKC subtypes, which plays a regulatory role in the increase of EOS in nasal polyps
