目的 从转录水平研究系统性红斑狼疮患者凋亡相关基因Fas、sFas表达情况 ,探索凋亡在SLE发病中的作用。方法 利用荧光标记RT PCR半定量方法 ,以 β actin为内参照 ,对 5 0例SLE、2 4例RA和 2 4名正常人外周血白细胞凋亡相关基因Fas、sFasmRNA表达水平进行了检测。结果 SLEFas、sFas表达均高于正常对照 ( 3 2± 1 8vs 2 1± 0 6,P <0 0 0 1和 1 2± 0 7vs 0 5 8±0 2 7,P <0 0 0 0 1) ,并且表达量与病情活动度相关 (相关系数分别为 0 5 2 18,P =0 0 0 7和 0 4 188,P =0 0 3 7) ,而RA仅sFas表达增高 ( 0 9± 0 5vs 0 5 8± 0 2 7,P <0 0 1)。结论 SLE患者外周血白细胞凋亡相关基因表达存在异常 ,提示凋亡在SLE发病中可能具有一定作用和意义。
Objective To study the expression of Fas and sFas in patients with systemic lupus erythematosus (SLE) at the transcriptional level and explore the role of apoptosis in the pathogenesis of SLE. Methods Semi-quantitative RT-PCR and β actin were used to detect the expression of Fas and sFas mRNA in peripheral blood mononuclear cells of 50 SLE patients, 24 RA patients and 24 normal controls. Results The expressions of SLEFas and sFas were significantly higher than those of the normal controls (32 ± 18 vs 21 ± 0 6, P <0 01 1 and 1 2 ± 0 7 vs 0 58 ± 0 2 7, P 0 01 0 1) , And the expression level correlated with the degree of disease activity (correlation coefficients were 0 5 2 18, P = 0 0 0 7 and 0 4 188 respectively, P = 0 0 3 7), while the expression of sFas increased only in RA (0 9 ± 0 5 vs 0 5 8 ± 0 2 7, P <0 0 1). Conclusion There is abnormal expression of apoptosis-related genes in peripheral leukocytes in SLE patients, suggesting that apoptosis may play a role in the pathogenesis of SLE.
