以CEA单克隆抗体C50 包被微孔板 ,异硫氰酸苄基二乙烯三胺四乙酸络合Eu3+ 标记C17单抗。采用平衡法建立CEA时间分辨荧光免疫分析 ,数据采用Log -Logit函数和四参数logit函数数据处理程序处理。方法的批内和批间CV分别为 2 .97%和 1.6 0 % ,平均回收率为10 1.98% ,灵敏度为 0 .2 0 μg/L ,可测范围为 2 .39~ 50 8.9μg/L ,ED50 为 6 0 .91± 0 .56 μg/L。本方法与AFP和CA12 5和CA153无交叉 ,与CA199有 3.2 2 %的交叉反应。Eu3+ 标记抗体于 - 30℃保存一年多 ,免疫反应性基本无损失 ,同批试剂连续 5个月分析结果稳定。与Wallac同类产品比较 ,结果吻合。
The CEA monoclonal antibody C50 was used to coat microtiter plate wells, and benzyldiethylenetriamine tetraacetic acid complexed with Eu3 + labeled C17 mAb. The equilibrium method was used to establish CEA time-resolved fluorescence immunoassay. The data was processed by Log-Logit function and four-parameter logit function data processing program.
