通过绞股蓝Gynostemma. pantaphyllum愈伤组织培养,系统观察不同前体的中间体和抑制剂类物质对愈伤组织生长和总皂甙积累的影响。结果表明,醋酸钠(25ppm)单独使用可提高其鲜重与干重生长速率,与氨硫脲协同可提高鲜重与干重生长速率及皂甙产率。香叶醇(0.01%)单独或与氨硫脲协同均可提高其皂甙产率。此外,绞股蓝愈伤组织生长曲线和总皂甙积累曲线显示,总皂甙积累峰期滞后于愈伤组织生长峰期4—8天。
Through the Gynostemma pantaphyllum callus culture, we systematically observed the effects of different precursor intermediates and inhibitors on callus growth and total saponin accumulation. The results showed that sodium acetate (25ppm) alone could increase its fresh weight and dry weight growth rate, synergistic with thiosemicarbazone could increase fresh weight and dry weight growth rate and saponin yield. Geraniol (0.01%) alone or with thiourea in synergy can increase its saponin yield. In addition, the growth curve of Gynostemma callus and the total saponin accumulation curve showed that the peak of total saponin accumulation lagged 4-8 days after the callus growth peak.
