本文介绍了何首乌片中蒽醌类成分的两种含量测定方法。一种方法是:何首乌片中的游离蒽醌先用甲醇提取,然后蒸去甲醇,再用氯仿提取,俟提取完全,将提取液中氯仿蒸去,残渣用0.5%醋酸镁——甲醇液显色,以1,8-二羟基蒽醌为标准,何首乌片中的结合蒽醌则用甲醇提取,然后蒸去甲醇,残渣溶于氯仿用5N硫酸水解,俟水解完全,分取氯仿层,用蒸馏水洗至中性,蒸去氯仿,残渣用0.5N醋酸镁——甲醇液显色,然后进行比色法测定。另一种方法用5%氢氧化钠—2%氢氧化铵显色,实验步骤见前文《何首乌的质量研究(Ⅰ)》。两种方法的实验结果表明:两法的平均值基本一致,但前法之色泽较后法更为稳定,且不受光线照射的影响。
This article describes two methods for determining the content of anthraquinones in Polygonum multiflorum. One method is: free Radon in Polygonum multiflorum extract first with methanol, then distill off methanol, extract with chloroform, extract with hydrazine completely, evaporate chloroform from the extract, and residue with 0.5% magnesium acetate-methanol In color, with 1,8-dihydroxyanthraquinone as the standard, the combination of Polygonum multiflorum tablets was extracted with methanol, and then methanol was distilled off. The residue was dissolved in chloroform and hydrolyzed with 5N sulfuric acid. The hydrazine was completely hydrolyzed and the chloroform layer was separated. Distilled water was washed to neutrality, chloroform was distilled off, the residue was developed with 0.5 N magnesium acetate-methanol, and then colorimetric assay was performed. Another method is to use 5% sodium hydroxide - 2% ammonium hydroxide color, the experimental steps see the previous “Quality Study of Polygonum multiflori (I)”. The experimental results of the two methods show that the average values of the two methods are basically the same, but the color of the former method is more stable than the latter method, and is not affected by light irradiation.
