目的研究重组脂联素(AD)腺相关病毒载体对大鼠滑膜成纤维细胞(SFLs)肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)和B细胞激活因子(BAFF)mRNA表达的影响。方法采用胶原酶消化法制备大鼠SFLs。携带AD基因的腺相关病毒按不同转染复数(MOI)转染大鼠SFLs,于第7天采用RT-PCR方法检测细胞因子AD、TNF-α、IL-6、BAFF mRNA表达。结果成功培养出大鼠SFLs,转染后的SFLs AD mRNA表达随MOI升高而增加;在MOI=5×105v.g./cell时,TNF-α、IL-6、BAFF mRNA表达最高。结论大鼠SFLs中高表达AD能够促进TNF-α、IL-6和BAFFmRNA表达,提示AD参与炎性反应和B细胞活化,在类风湿关节炎的慢性炎症过程中发挥重要作用。
Objective To investigate the effect of recombinant adeno-associated virus vector of adiponectin on the expression of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) and B-cell activating factor (BAFF) in synovial fibroblasts (SFLs) ) mRNA expression. Methods Rat SFLs were prepared by collagenase digestion. The adeno-associated virus carrying AD gene was transfected into rat SFLs at different MOIs, and the expression of cytokines such as AD, TNF-α, IL-6 and BAFF mRNA was detected by RT-PCR on the 7th day. Results SFLs were successfully cultured and the expression of AD mRNA in SFLs was increased with the increase of MOI. The highest expression of TNF-α, IL-6 and BAFF mRNA was observed at MOI = 5 × 105v.g./cell. Conclusion High expression of AD in rat SFLs can promote the expression of TNF-α, IL-6 and BAFFmRNA, suggesting that AD is involved in the inflammatory response and B cell activation and plays an important role in the chronic inflammatory process of rheumatoid arthritis.
