目的观察单纯疱疹病毒胸苷激酶(HSV-TK)丙氧鸟苷(GCV)自杀基因系统联合热疗(hyperthermia)对人肝癌细胞系HepG2体外的杀伤作用。方法采用脂质体转染法将HSV-TK基因转染HepG2细胞,观察GCV联合热疗对HepG2/tk的体外杀伤作用。结果热疗与GCV联用组各温度作用下细胞凋亡率均较对照组和单用GCV组明显升高,差异均有统计学意义(均P<0.05)。随着温度的升高,热疗联合GCV组诱导细胞凋亡的效果也增强,44℃时细胞增殖抑制率为(78.8±7.5)%,细胞凋亡率达(42.8±3.6)%。结论脂质体可介导HSV-TK基因转染入HepG2细胞并获得稳定表达,热疗可显著提高GCV对HepG2/tk细胞的杀伤效果。
Objective To observe the killing effect of HSV-TK gonorrhoea (GCV) suicide gene system combined with hyperthermia on human hepatoma cell line HepG2 in vitro. Methods HSV-TK gene was transfected into HepG2 cells by lipofection. The killing effect of GCV combined with hyperthermia on HepG2 / tk in vitro was observed. Results Compared with the control group and GCV group, the apoptosis rates of hyperthermia group and GCV group were significantly increased (all P <0.05). With the increase of temperature, the effect of hyperthermia combined with GCV on apoptosis was also enhanced. The rate of cell proliferation was (78.8 ± 7.5)% at 44 ℃, and the apoptosis rate was (42.8 ± 3.6)% at 44 ℃. Conclusion The liposome can mediate the transfection of HSV-TK gene into HepG2 cells and achieve stable expression. Hyperthermia can significantly increase the killing effect of GCV on HepG2 / tk cells.
