目的:通过对超氧化物岐化酶( S O D)模拟物原儿茶醛缩氨基硫脲铜(Ⅱ)配合物(protocatechualdehydethiosem icarbazonecopper(Ⅱ)com plexs, P T C)清除 O2·自由基及其生物学作用的实验观察,从自由基清除剂的角度寻求开发高效低毒的中药有效成份金属配合物新药. 方法:①以黄嘌呤黄嘌呤氧化酶 Lum inol化学发光体系测定 P T C的 S O D样活性. ②实验观察 P T C对激活的人白细胞吞噬发光、血液流变性、脂质过氧化产物的影响和对烧伤后继发氧化损伤的保护作用. 结果:① P T C 具有很高的 S O D 样活性. 其[ I D]50 为150 μg/ L,是 S O D([ I D]50 为300 μg/ L)的2 倍. ② P T C能显著地抑制酵母多糖激活的人白细胞的吞噬发光. ③ P T C能降低由于白细胞的激活而引起的人血液粘度和脂质过氧化产物( M D A)的升高. ④ P T C能使小鼠烧伤模型的炎症反应减轻,并能使烧伤小鼠血清中增高的 M D A 和一氧化氮代谢产物( N O2 ·)显著降低. 结论: P T C是一种很强的 O2·清除剂,对 O2·介导的氧化损伤有保护作用.
OBJECTIVE: To investigate the effects of protocatechualdehydethiosem icarbazoneopper (Ⅱ) plexs (P Tc) on the activity of superoxide dismutase (S O D) Clear O2 · free radicals and their biological effects of experimental observation, from the perspective of free radical scavengers to develop efficient and low toxicity of traditional Chinese medicine active ingredient metal complexes of new drugs. Methods: ① xanthine xanthine oxidase Lum inol chemiluminescence system for determination of P T C S D-like activity. ② The effect of P T C on activated leukocyte phagocytosis, hemorheology and lipid peroxidation and the protective effect on secondary oxidative damage after burn were observed. Results: ① P T C had high S O D-like activity. Its [I D] 50 is 150 μg / L, which is double that of S O D ([I D] 50 300 μg / L). ② P T C can significantly inhibit the phagocytosis of human leukocyte activated by zymosan. ③ P T C can reduce the human blood viscosity and the increase of lipid peroxidation product (M D A) caused by the activation of leukocytes. ④ P T C can reduce the inflammatory reaction in mouse burn model and decrease the serum M D A and NO (N O 2 · ) in the serum of burned mice. CONCLUSION: P T C is a strong O2 scavenger, which has a protective effect on O2 mediated oxidative damage.
