Objective: To investigate the effect of tea polyphenols on the growth of prostate cancer cells and its mechanism. Methods: The hormone-independent prostate cancer cell line DU145 was selected as the research object. Tea polyphenols were added into the drug group to make the final concentrations of 50, 100, 250 and 500 μg / ml respectively. The control group was added with normal cell culture medium . After cultured for 48 h in each group, cell viability was detected by MTT colorimetric assay. Survivin gene expression in each group was detected by Western blot analysis and quantitative RT-PCR. RESULTS: The cell viability of each drug group was 0.97 ± 0.12, 0.71 ± 0.07, 0.20 ± 0.03 and 0.08 ± 0.01, respectively, 48 h after the addition of tea polyphenol solution. Compared with the control group, ), The other three drug group cell survival rates were significantly lower than the control group (P <0.01). With the increase of the time of tea polyphenols, the cell survival rate of all the groups showed a downward trend. By 96 h, the survival rates of the other three groups were below 5% except the 50 μg / ml group. After 48 h, the gray values of sur-vivin in control, 100, 250 and 500 μg / ml groups were 15 075 ± 48, 13 425 ± 31, 2 017 ± 24, 1 274 ± 22, Compared with the control group, the expression of survivin protein in the three drug groups were significantly decreased (all P <0.01). In addition, the amounts of survivin mRNA in the groups of 50,100,250 and 500μg / ml for 48 h were 0.74 ± 0.03,0.64 ± 0.02,0.52 ± 0.01 and 0.21 ± 0.02, respectively, which were significantly lower than those in the control group (P < 0.01). Conclusion: Tea polyphenols can inhibit the growth of prostate cancer DU145 cells, and this effect may be related to the reduction of survivin gene expression.
