目的 为了观察白细胞介素6(IL-6)反义RNA对肾小球系膜细胞(MC)的影响。方法 将IL-6反义RNA用脂质体导入包装细胞PA_(317),经C_(418)筛选,PCR技术分析,并用~3H-胸腺嘧啶(~3H-TdR)掺入法、KD_(83)细胞株、North-ern杂交技术分别检测IL-6反义RNA对肾小球系膜细胞增殖、分泌IL-6以及表达IL-6mRNA的影响。结果 重组假病毒滴度为1×10~7CFU/ml,肾小球系膜细胞~3H-TdR掺入率及IL-6产生与mRNA表达均明显低于对照组。结论 IL-6反义RNA可抑制MC增殖,下调MC中IL-6的产生与表达。 Objective To investigate the effects of interleukin-6 (IL-6) antisense RNA on mesangial cells (MC). Methods IL - 6 antisense RNA was introduced into the packaging cell line PA 317 by lipofectamine 2000. The recombinant plasmid was identified by C_ (418) screening and PCR analysis. The expression of KD_ (83) was detected by ~ 3H thymidine incorporation ) Cell line and North-ern hybridization technique were used to detect the effects of IL-6 antisense RNA on the proliferation, secretion of IL-6 and IL-6 mRNA expression in glomerular mesangial cells. Results The titer of recombinant pseudovirus was 1 × 10-7 CFU / ml, the incorporation rate of 3H-TdR and the production of IL-6 and the mRNA expression of mesangial cells in glomerular mesangial cells were significantly lower than those in control group. Conclusion IL-6 antisense RNA can inhibit the proliferation of MC and down-regulate the production and expression of IL-6 in MC.
