目的:观察不同碘缺乏大鼠肾组织UCP2、caspase-9蛋白的表达,探讨二者表达的相关性。方法:雌性Wistar大鼠随机分为轻度缺碘组、重度缺碘组及正常对照组,总碘摄入量分别为5、1.24、10μg/d,采用RT-PCR、免疫组化等方法检测大鼠离体肾脏UCP2及caspase-9蛋白的表达,并分析二者表达的相关性。结果:与正常对照组相比,碘缺乏大鼠肾组织UCP2基因、蛋白表达水平下降,caspase-9蛋白表达增加(P<0.05),远端肾小管、髓质肾小管改变更明显;且caspase-9蛋白表达与UCP2表达负相关。结论:碘缺乏导致大鼠肾组织UCP2表达下降,可能与凋亡蛋白caspase-9表达增加有关。
OBJECTIVE: To observe the expression of UCP2 and caspase-9 in renal tissue of different iodine deficiency rats, and to explore the correlation between them. Methods: Female Wistar rats were randomly divided into mild iodine deficiency group, severe iodine deficiency group and normal control group. The total iodine intake was 5, 1.24 and 10 μg / d, respectively. RT-PCR and immunohistochemistry The expression of UCP2 and caspase-9 in isolated rat kidney and the correlation between them were analyzed. Results: Compared with the normal control group, the UCP2 gene expression in the kidney of iodine-deficient rats decreased, the expression of caspase-9 protein increased (P <0.05), and the changes of distal tubular and medullary tubules were more obvious. -9 protein was negatively correlated with UCP2 expression. Conclusion: Iodine deficiency leads to the decrease of UCP2 expression in rat kidney, which may be related to the increase of caspase-9 expression.
