目的:以Hep G2.2.15细胞系为体外实验模型,研究波棱素脂质体体外抑制乙肝病毒的生物学活性。方法:MTT法检测药物对细胞的毒性,将不同安全浓度的波棱素脂质体及阳性对照药物拉米夫定(3TC)作用于此细胞系,同时实验设不加药物的细胞对照,给药3,6d后收集细胞培养上清液。采用ELISA法测定各组细胞培养上清液中HBs Ag,HBsAg水平,实时定量PCR检测HBV-DNA的含量。结果:波棱素脂质体对HepG2.2.15细胞的TC50为1443.68μg/ml。波棱素脂质体对HepG2.2.15分泌的HBs Ag和HBeAg的IC50分别为131.08μg/ml,109.50μg/ml,治疗指数为11.01和13.18。同时,浓度范围在25~200μg/ml时明显抑制HBV-DNA表达,最高抑制率达到55.04%。结论:波棱素脂质体具有一定的体外抑制乙肝病毒复制及抗原表达的作用。
OBJECTIVE: To study the inhibitory effect of ribonin liposomes on the biological activity of hepatitis B virus in Hep G2.2.15 cell line in vitro. Methods: MTT assay was used to determine the cytotoxicity of the drug. The cells were treated with lamivudine (3TC) and lipopolysaccharide (LPS) at different safe concentrations for 3 days, After 3 and 6 days, the cell culture supernatants were collected. The levels of HBsAg and HBsAg in the cell culture supernatants of each group were measured by ELISA, and the content of HBV-DNA was detected by real-time quantitative PCR. Results: The TC50 of ribonin liposome on HepG2.2.15 cells was 1443.68μg / ml. The IC50 of ribonin liposomes on HBsAg and HBeAg secreted by HepG2.2.15 were 131.08μg / ml and 109.50μg / ml respectively, and the therapeutic indexes were 11.01 and 13.18. At the same time, HBV-DNA expression was significantly inhibited at the concentration range of 25 ~ 200μg / ml with the highest inhibition rate of 55.04%. Conclusion: The ribonin liposomes have a certain inhibitory effect on hepatitis B virus replication and antigen expression in vitro.
