本试验选用内蒙古科尔沁草甸草原野生大豆(父本)与当地栽培大豆品种大白眉(母本)杂交后代的稳定株系9002的下胚轴作为供试外植体,探讨了大豆种子灭菌时间、激素类型等因素对大豆继代培养的影响。结果表明:①该大豆种子的最佳灭菌条件是:先70﹪乙醇杀菌1min,然后6﹪NaClO灭菌17min。②大豆诱导不定芽发育适宜培养基:MS+6-BA0.5 mg/L+KT1.0mg/L+IAA0.5 mg/L+蔗糖3﹪+琼脂0.8﹪。③适宜绿芽分化生根培养基:MS+NAA0.5 mg/L+蔗糖2﹪+琼脂0.8﹪为培养基。对大豆进行组织培养技术的成功有利于对加速良种繁育、优良种质资源保存、有益突变体筛选及大豆遗传操作等都具有重要意义。
In this study, the hypocotyls of the stable strain 9002 from the wild soybean (male parent) of Horqin meadow grassland in Inner Mongolia and the hybrid cultivar Dahongmei (female parent) of the local cultivated soybean were used as the explants to investigate the effects of soybean seed sterilization time , Hormone type and other factors on the subsequent subculture of soybean. The results showed that: ① The best sterilization conditions of the soybean seeds were as follows: first 70% ethanol sterilization 1min, then 6% NaClO sterilization 17min. ② Soybean induced adventitious bud development suitable medium: MS + 6-BA0.5 mg / L + KT1.0 mg / L + IAA0.5 mg / L + sucrose3% + agar0.8%. ③ Suitable rooting culture medium differentiation: MS + NAA0.5 mg / L + 2% sucrose + 0.8% agar medium. The success of tissue culture techniques for soybeans is of great importance for accelerating seed breeding, preservation of good germplasm, selection of useful mutants, and genetic manipulation of soybeans.
