目的探讨急性百草枯染毒大鼠肺组织内Th17细胞相关细胞因子的表达变化及IL-17抗体的中和作用。方法将72只健康SPF级Wistar大鼠随机分为对照(生理盐水)组、百草枯中毒组和抗体中和组,每组24只,雌雄各半。百草枯中毒组和抗体中和组一次性灌胃染毒百草枯(50mg/kg);抗体中和组2 h后腹腔注射3μg/ml的IL-17抗体。分别于染毒后第1、3、7、21天,检测外周血IL-17、IL-6、TGF-β浓度和肺组织中维甲酸孤独核受体γt(RORγt)mRNA的表达水平。观察肺组织的病理学改变。结果与对照组相比,百草枯中毒组和抗体中和组雌、雄大鼠各时间点血清IL-17、IL-6、TGF-β浓度及肺组织RORγtmRNA的表达均升高,差异有统计学意义(P<0.05);与百草枯中毒组相比,抗体中和组雌、雄大鼠第1、3天血清IL-17、IL-6含量及第3、7天血清TGF-β含量以及第1、3、7天肺组织RORγt mRNA表达水平均降低,差异有统计学意义(P<0.05)。而雄性大鼠第7天血清IL-17含量及雌性大鼠第21天肺组织RORγt mRNA表达水平明显降低,差异有统计学意义(P<0.05)。结论 Th17细胞参与了百草枯中毒大鼠急性炎症反应过程,IL-17抗体对急性百草枯致肺损伤具有拮抗作用。
Objective To investigate the changes of Th17 cell-related cytokines and the neutralization of IL-17 in the lung of acute paraquat-induced rats. Methods Seventy-two healthy Wistar rats were randomly divided into control (normal saline) group, paraquat poisoning group and antibody neutralization group, 24 rats in each group, with half male and one female. The paraquat poisoning group and the antibody neutralization group were treated with paraquat (50mg / kg) by gavage once a day, while the antibody-neutralizing group was injected intraperitoneally with 3μg / ml IL-17 antibody 2h later. The concentrations of IL-17, IL-6 and TGF-β in the peripheral blood and the expression of RORγt mRNA in the lung tissue were detected on the 1st, 3rd, 7th and 21st day after exposure respectively. Observe the pathological changes of lung tissue. Results Compared with the control group, the levels of IL-17, IL-6, TGF-β and the expression of RORγt mRNA in the paraquat poisoning group and the antibody neutralizing group at each time point were significantly increased (P <0.05). Compared with the paraquat poisoning group, the levels of IL-17 and IL-6 in serum of the male and female rats on the 1st and 3rd day after antibody neutralization and the levels of TGF- The mRNA expression of RORγt in lungs decreased on the 1st, 7th and 7th day (P <0.05). However, the level of IL-17 in serum on the 7th day in male rats and the expression of RORγt mRNA in lungs on the 21st day in the female rats were significantly decreased (P <0.05). Conclusion Th17 cells are involved in the process of acute inflammation in paraquat poisoning rats. IL-17 antibody has an antagonistic effect on acute lung injury induced by paraquat.
