作者应用膜片钳吸附式构型和内面向外式构型研究了麻醉药乙托咪酯对乳大鼠大脑皮质感觉区锥体神经元钙激活钾通道(Kca)的影响。取生后1-3天SD乳大鼠大脑皮质感觉区,机械吹打游离脑细胞,接种培养24小时后,加入100μM阿糖胞苷以抑制非神经元的分裂增殖。选用培养5-7天结构典型的锥体神经元进行实验,研究结果表明:1、乳大鼠感觉皮质锥体神经元上存在有164±11PS的Kca通道(n29)。2、在吸附式构型时,三个不同浓度(0.8μM、4pM、20pM)的乙托咪酯明显激光锥体神经元的Kca通道,其开放概率明显呈浓度依赖性改变(Pwto01,n—3),对电流幅值影响不明显。3、在内面向外构型时,通道亦是明显激活。4、两种构型在外suM乙托咪酯作用时无明显差异(P>0-05)。5、钠酪酮对乙托咪醋直接作用于神经细胞膜离子通道,而钠酪酮可逆转乙托咪酯所致麻醉作用。
In this study, we investigated the effect of anesthetic ethanol ethyl ester on calcium-activated potassium channels (KCa) in the sensory pyramidal neurons of the cerebral cortex in the rat by using the patch-clamp adsorption configuration and the inner-facing outward configuration. After 1 to 3 days from birth, SD rats were sensitized to the cerebral cortex by mechanical beating of free brain cells. After 24 hours of inoculation, 100 μM of cytarabine was added to inhibit the proliferation and proliferation of non-neurons. The results showed that there were 164 ± 11 KCa channels (n29) in sensory cortical pyramidal neurons of the rats. (2) In the adsorbed configuration, the Kca channel of three distinct concentrations (0.8μM, 4pM, 20pM) of etomidate clearly showed a concentration-dependent change in the openness of Kca channels (Pwto01, n -3), the impact on the current amplitude is not obvious. 3, in the inside out configuration, the channel is also significantly activated. 4. There was no significant difference between the two configurations when using suM betaetomidate (P> 0-05). 5, sodium caseinate on the Bituo Mito directly on the membrane ion channels of nerve cells, and sodium casemate can reverse the anesthetic effects induced by etomidate.
