[目的]通过观察中药咳喘方对哮喘豚鼠肺组织嗜酸细胞凋亡的影响 ,进一步探讨中药咳喘方治疗哮喘的疗效机制。[方法]采用经典的哮喘豚鼠模型 ,将60只符合实验标准的雄性豚鼠随机分为4组 ,即正常对照组、模型组、泼尼松组和咳喘方组 ,镜下观察各组支气管肺泡灌洗液嗜酸细胞计数的变化及肺组织嗜酸细胞浸润和凋亡的情况 ,并采用免疫组化的方法观察各组肺组织嗜酸细胞Fas和bcl-2凋亡基因的表达情况 ,将结果进行统计学分析。[结果]咳喘方组支气管肺泡灌洗液嗜酸细胞计数和肺组织嗜酸细胞浸润的数量明显低于模型组和泼尼松组 ,而肺组织嗜酸细胞凋亡的数量则高于模型组和泼尼松组 ;咳喘方组肺组织嗜酸细胞凋亡基因Fas的表达较其他组增强 ,而凋亡基因bcl-2的表达较泼尼松组和模型组低。[结论]中药咳喘方能减少哮喘豚鼠肺组织嗜酸细胞的浸润 ,促进嗜酸细胞凋亡基因Fas的表达 ,诱导嗜酸细胞凋亡 ,这可能是其治疗哮喘的疗效机制之一。
[Objective] To observe the effect of Kechuan recipe on the apoptosis of eosinophils in asthmatic guinea pigs, and further explore the therapeutic mechanism of traditional Chinese medicine Kechuan recipe on asthma. [Methods] Using the classic asthma guinea pig model, 60 male guinea pigs meeting the experimental criteria were randomly divided into 4 groups: normal control group, model group, prednisone group, and Kechuan Fang group. Bronchoalveolar bronchoalveolar bronchoalveolar bronchoalveolar bronchoalveolar lavage was observed under microscope. The change of eosinophil count in lavage fluid and eosinophil infiltration and apoptosis in lung tissue were observed. Immunohistochemistry was used to observe the expression of Fas and bcl-2 apoptotic genes in lung tissue of each group. The results were analyzed statistically. [Results] The number of eosinophils in the bronchoalveolar lavage fluid and the amount of eosinophil infiltration in the lungs in the Kechuan Fang group were significantly lower than those in the model group and the prednisone group, while the amount of eosinophil apoptosis in the lung was higher than that in the model. The expression of Fas in the lung tissue of the Kechuanfang group was stronger than that of the other groups, but the expression of bcl-2 was lower in the prednisone group and the model group than in the prednisone group. [Conclusion] Kechuanfang can reduce the infiltration of eosinophils in lung tissue of asthmatic guinea pigs, promote the expression of Fas gene and induce apoptosis of eosinophils in asthmatic guinea pigs, which may be one of the therapeutic mechanisms for the treatment of asthma.
