目的 观察局灶性脑缺血对 N -甲基 - D-天门冬氨酸受体 (NMDAR)及 NMDAR m RNA表达的影响。方法 采用线栓法制作可复流大脑中动脉闭塞大鼠模型。 30只雄性大鼠随机分为 3组 :正常组、假手术组和大脑中动脉闭塞 (MCAO)组。 MCAO后第 5天处死动物 ,取脑组织进行 NR1 免疫组化及 NR1 m RNA原位杂交检测。结果 (1)正常组海马各区均有 NR1 阳性细胞的分布 ,其中海马 CA3 区、DG区分布较多 ;MCAO组 NR1 阳性细胞在海马 CA1 区、CA3区及 DG区的数密度、光密度较正常组高 ;(2 )正常组海马各区均有极少量的 NR1 m RNA阳性细胞分布 ;MCAO组 NR1 m RNA阳性细胞在海马 CA1 区、CA3 区的数密度、光密度均较正常组有显著性增高。结论 大鼠海马各区都存在着 NR1 及 NR1 m RNA不同程度的表达 ;MCAO后 NR1 及 NR1 m RNA的表达水平上调。
Objective To observe the effect of focal cerebral ischemia on the expression of N-methyl-D-aspartate receptor (NMDAR) and NMDAR m RNA. Methods The rat model of re-flow middle cerebral artery occlusion was made by thread plug method. Thirty male rats were randomly divided into three groups: normal group, sham operation group and middle cerebral artery occlusion (MCAO) group. Animals were sacrificed on the fifth day after MCAO, and brain tissues were harvested for NR1 immunohistochemistry and NR1 m RNA in situ hybridization. Results (1) The distribution of NR1 positive cells in the normal hippocampus of all groups was higher than that in the normal control group, especially in the CA3 and DG regions of the hippocampus. The numbers density and optical density of NR1 positive cells in hippocampus CA1, CA3 and DG were more than normal (2) There were very few NR1 m RNA positive cells in normal hippocampus. The number density and optical density of NR1 m RNA positive cells in hippocampus CA1 and CA3 in MCAO group were significantly higher than those in normal group . Conclusion NR1 and NR1 m RNA are all expressed in different regions of rat hippocampus. The expressions of NR1 and NR1 m RNA are up-regulated after MCAO.
