目的:探讨葛根素对肺缺血-再灌注损伤(PIR I)时Fas/FasL表达的影响。方法:采用在体兔单肺原位缺血-再灌注模型。实验兔70只,随机分为假手术对照组(sham,10只)、肺缺血-再灌注组(I-R,30只)和肺缺血-再灌注加葛根素组(LGT,30只)。每组又分为再灌注1 h、3 h、5 h 3个亚组,每个亚组10只,分别于再灌注1 h、3 h、5 h 3个时点取左肺组织,观察Fas/Fas配体(Fas/FasL)mRNA定位表达、凋亡指数(AI)、肺组织湿干重比(W/D)、肺损伤组织学定量评价指标(IQA)及光镜、电镜下的组织形态学改变。结果:肺再灌注1 h、3 h、5 h,LGT组Fas/FasL mRNA在肺小动脉内(外)膜、肺小静脉内膜、肺泡上皮及肺支气管上皮呈弱阳性表达,明显低于同一时点I-R组(P<0.05);AI、W/D和IQA值显著低于I-R组(P<0.01和P<0.05);肺组织形态学异常改变不同程度减轻。结论:葛根素可下调肺组织Fas/FasL mRNA的表达而减轻细胞凋亡,对PIR I发挥积极的防治作用。
Objective: To investigate the effect of puerarin on the expression of Fas/FasL in lung ischemia-reperfusion injury (PIR I). METHODS: A single lung in situ ischemia-reperfusion model in rabbits was used. Seventy rabbits were randomly divided into sham-operated control group (sham, 10), pulmonary ischemia-reperfusion group (I-R, 30), and lung ischemia-reperfusion plus puerarin group (LGT, 30). Each group was divided into three subgroups: reperfusion 1 h, 3 h, and 5 h. Each subgroup was divided into 10 groups. Left lung tissue was taken at 1 h, 3 h, and 5 h after reperfusion, and Fas was observed. Fas/FasL mRNA expression, apoptosis index (AI), wet/dry weight ratio (W/D) of lung tissue, quantitative assessment index (IQA) of lung injury, light microscopy and electron microscopy Morphological changes. RESULTS: At 1 h, 3 h, and 5 h after reperfusion, LPS showed weakly positive Fas/FasL mRNA expression in the pulmonary arteriole (outer) membrane, small intrapulmonary vein, alveolar epithelium, and lung bronchial epithelium. At the same time, IR group (P<0.05); AI, W/D and IQA values were significantly lower than IR group (P<0.01 and P<0.05); abnormal changes of lung morphological changes were alleviated to some extent. Conclusion: Puerarin can down-regulate the expression of Fas/FasL mRNA in lung tissue and reduce apoptosis. It plays an active role in prevention and treatment of PIR I.
