目的:研究胡颓子叶正丁醇萃取部位的化学成分及其对人宫颈癌、人胃癌细胞的细胞毒活性。方法:采用反复柱色谱方法进行分离,利用化合物的光谱学数据结合理化性质鉴定化合物的结构;采用MTT法测定化合物对体外肿瘤传代细胞SGC-7901(人胃癌)和Hela(人宫颈癌)的细胞毒活性。结果:分离并鉴定了4个化合物:3′-甲氧基槲皮素(Ⅰ)、山柰酚-3-O-β-D-6-O-(对羟基桂皮酰基)-吡喃葡萄糖苷(Ⅱ)、葛花苷(Ⅲ)、3′-O-甲基鞣花酸-4-O-α-L-吡喃鼠李糖苷(Ⅳ)。4个化合物对体外肿瘤传代细胞SGC-7901(人胃癌)和Hela(人宫颈癌)有一定的抑制作用。结论:化合物Ⅰ、Ⅲ、Ⅳ为首次从该种植物中分离得到。化合物Ⅰ和Ⅱ对Hela细胞株有明显的细胞毒作用。
OBJECTIVE: To study the chemical constituents of n-butanol-extracted parts of Elaeagnus and their cytotoxic activities on human cervical cancer and human gastric cancer cells. METHODS: The compounds were separated by repeated column chromatography. The structures of the compounds were identified by spectroscopic data and physical and chemical properties of the compounds. The compounds were tested by MTT assay for the cells of tumor-bearing cells SGC-7901 (human gastric cancer) and Hela (human cervical cancer) Toxic activity. RESULTS: Four compounds were isolated and identified: 3’-methoxyquercetin (Ⅰ), kaempferol-3-O-β-D-6-0- (p-hydroxy cinnamoyl) -glucopyranoside (Ⅱ), kaempferide (Ⅲ) and 3’-O-methyl-ellagic acid-4-O-α-L-rhamnopyranoside (Ⅳ). Four compounds have some inhibitory effects on tumor cell line SGC-7901 (human gastric cancer) and Hela (human cervical cancer) in vitro. Conclusion: Compounds Ⅰ, Ⅲ and Ⅳ were isolated from this plant for the first time. Compound Ⅰ and Ⅱ have obvious cytotoxicity on Hela cell line.
