目的观察参地颗粒对系膜增生性肾小球肾炎(Ms PGN)大鼠T细胞亚群及足细胞标志蛋白的影响,探讨参地颗粒的作用机制。方法建立Ms PGN模型,将40只SD大鼠随机分为模型组、缬沙坦组、参地颗粒组和正常组。参地颗粒组按4 g/(kg·d)灌胃,缬沙坦组按10.3 mg/(kg·d)灌胃,模型组及正常组每日给予等量温水灌胃,连续灌胃12周。常规生化法检测各组大鼠24 h尿蛋白定量、流式细胞术检测全血CD4+T细胞、CD8+T细胞数量,ELISA检测血清白细胞介素2(IL-2)、IL-4、IL-17和尿足萼糖蛋白(PCX)、B7-1水平及肾脏组织中钙调磷酸酶(Ca N)含量。结果模型组24 h尿蛋白定量、CD8+T细胞水平,血清IL-2、IL-17和尿PCX、B7-1水平及肾脏组织中Ca N含量均高于正常组;缬沙坦组和参地颗粒组以上指标均低于模型组,参地颗粒组优于缬沙坦组。模型组全血CD4+T细胞及血清IL-4水平均低于正常组,缬沙坦组和参地颗粒组以上指标均高于模型组,参地颗粒组优于缬沙坦组。结论参地颗粒可有效降低Ms PGN大鼠24 h尿蛋白定量,可能与增加CD4+T细胞数量,降低血清IL-2、IL-17及尿PCX、B7-1水平和肾脏组织中Ca N的表达,增加血清IL-4水平有关。
Objective To observe the effect of Shen Di Granule on T cell subsets and podocyte proteins in mesangial proliferative glomerulonephritis (Ms PGN) rats and to explore the mechanism of Shen Di Granule. Methods Ms PGN model was established. 40 SD rats were randomly divided into model group, valsartan group, Shendi Granule group and normal group. The levels of CD4 + T cells and serum IL-4 in the model group were lower than those in the normal group. Conclusion Shen-gong Granule can effectively reduce 24-h urinary protein in Ms PGN rats, which may be associated with increasing the number of CD4 + T cells, decreasing the levels of IL-2, IL-17 and urinary PCX, B7-1 and Ca N in kidney Expression, increased serum IL-4 levels.
