目的 探讨h Spry2基因在 13 6例肿瘤发生中的作用。 方法 分别采用逆转录 聚合酶链式反应 (RT PCR)、聚合酶链式反应 (PCR)、单链构象多态性分析 (SSCP)和DNA测序技术检测h Spry2基因在部分人类肿瘤及肿瘤细胞系中的表达、缺失和突变。 结果 h Spry2基因在 71例癌肿瘤及瘤旁正常组织中均有表达 ,表达差异无显著性无定量分析 ,故无统计学处理。该基因在65例骨及软组织肿瘤中无纯合性缺失。在MKN45细胞系cDNA中存在 15处同义突变 ;在DLD 1细胞系cDNA和基因组DNA中存在 1处相同的同义突变。h Spry2基因与果蝇Sprouty同源序列的扩增片段等长于用相同引物扩增的cDNA片段长度。结论 h Spry2基因在上述肿瘤的发生中不起作用 ;h Spry2基因与果蝇Sprouty同源序列中不含有内含子。
Objective To investigate the role of hspry2 gene in 136 cases of tumorigenesis. Methods Reverse transcription polymerase chain reaction (RT PCR), polymerase chain reaction (PCR), single strand conformation polymorphism (SSCP) and DNA sequencing were used to detect the expression of hspry2 gene in some human tumors and tumor cell lines In the expression, deletion and mutation. Results The hspry2 gene was expressed in 71 cases of cancer and adjacent normal tissues. There was no significant difference between the two groups. There was no statistical significance. The gene in 65 cases of bone and soft tissue tumors without homozygous deletion. There are 15 synonymous mutations in the MKN45 cell line cDNA; 1 synonymous mutation exists in the DLD 1 cell line cDNA and genomic DNA. h Spry2 gene and Drosophila sprouty homologous sequence amplified fragment length equal to the length of cDNA fragments amplified with the same primers. Conclusion The hspry2 gene has no effect on the above tumor occurrence. The hspry2 gene does not contain introns in Sprouty homologue of Drosophila.
