目的探讨人骨髓来源间充质干细胞(BMSCs)诱导分化成软骨细胞过程中协同刺激分子的表达及其对细胞免疫原性改变的影响。方法用密度梯度离心法分离骨髓,体外培养BMSCs,经形态学观察、流式细胞仪鉴定后诱导成软骨样细胞,用流式检测细胞表面协同刺激分子的表达情况;将未分化的BMSCs及诱导后的成软骨样细胞分别与T细胞共培养,采用3H-TdR法检测T细胞的增殖情况。结果人BMSCs不表达或低表达协同刺激分子CD28、CD80、CD83、CD86,抑制T细胞增殖;而诱导后的成软骨细胞上调表达CD28、CD80、CD83、CD86,并促进T细胞增殖。结论人BMSCs对T细胞增殖有抑制作用;而诱导后的成软骨细胞免疫原性增强,促进T细胞增殖。
Objective To investigate the expression of costimulatory molecules during the differentiation of human bone marrow-derived mesenchymal stem cells (BMSCs) into chondrocytes and their effects on the changes of cellular immunogenicity. Methods BMSCs were isolated by density gradient centrifugation. BMSCs were cultured in vitro. After morphological observation and flow cytometry, BMSCs were induced to differentiate into chondrocyte-like cells. The expression of co-stimulatory molecules was detected by flow cytometry. After the chondrocyte-like cells were co-cultured with T cells, 3H-TdR method was used to detect the proliferation of T cells. Results Human BMSCs did not express or co-stimulate CD28, CD80, CD83 and CD86, and inhibited the proliferation of T cells. However, the induced chondrocytes up-regulated the expression of CD28, CD80, CD83 and CD86, and promoted the proliferation of T cells. Conclusion Human BMSCs can inhibit the proliferation of T cells. However, the induced chondroblasts can enhance the immunogenicity and promote the proliferation of T cells.
