目的探讨Tribble3(TRIB3)在高糖促进心肌成纤维细胞合成Ⅰ型胶原中的作用。方法体外培养大鼠心肌成纤维细胞,采用实时定量逆转录一多聚酶链反应和酶标记免疫吸附测定法分别测定TRIB3mRNA及Ⅰ型前胶原的mRNA和蛋白的表达,化学合成针对大鼠TRIB3的siRNA,转染大鼠心肌成纤维细胞,观察高糖对心肌成纤维细胞合成Ⅰ型前胶原的变化。结果高糖不仅促进心肌成纤维细胞Ⅰ型胶原mRNA和蛋白质表达,而且增加TRIB3mRNA的表达(P<0.01);TRIB3siRNA抑制TRIB3的表达后,高糖促进Ⅰ型胶原mRNA和蛋白质表达的作用下降(P均<0.01)。结论 TRIB3参与了高糖促进心肌成纤维细胞Ⅰ型胶原合成。
Objective To investigate the role of Tribble3 (TRIB3) in the synthesis of type Ⅰ collagen induced by high glucose in cardiac fibroblasts. Methods Rat cardiac fibroblasts were cultured in vitro. The mRNA and protein expressions of TRIB3 mRNA and type Ⅰ procollagen were determined by real-time quantitative reverse transcription-polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. The siRNA targeting TRIB3 of rat was chemically synthesized, Transfection of rat cardiac fibroblasts, to observe the changes of type Ⅰ procollagen induced by high glucose in cardiac fibroblasts. Results High glucose not only promoted the mRNA and protein expression of type Ⅰ collagen but also increased the expression of TRIB3 mRNA in cardiac fibroblasts (P <0.01). After TRIB3 siRNA inhibited the expression of TRIB3, the effect of high glucose on the expression of type Ⅰ collagen mRNA and protein decreased All <0.01). Conclusion TRIB3 is involved in the high glucose induced type Ⅰ collagen synthesis in cardiac fibroblasts.
