目的研究化痰活血方(Hua-Tan-Huo-Xue-Fang,HTHXF)对卵清蛋白(OVA)引起的哮喘小鼠肺泡灌洗液(Bronchoalveolar lavage fluids,BALF)中白细胞数目及IL-5、IL-10、TGF-β1、MUC5AC表达的影响,并探究其治疗哮喘的可能作用机制。方法腹腔注射含有OVA与氢氧化铝的致敏液致敏,雾化吸入OVA激发哮喘模型。于实验第9天灌胃给药,雾化激发期间灌胃时间控制在雾化激发前1h。实验第28天处死小鼠,并取BALF测定其中的白细胞数目及IL-5、IL-10、TGF-β1和MUC5AC水平。结果化痰活血方能明显降低BALF中淋巴细胞、中性粒细胞、嗜酸性粒细胞、单核细胞和白细胞总数,降低IL-5和MUC5AC的水平,升高IL-10和TGF-β1水平。结论化痰活血方治疗哮喘的作用机制可能与化痰活血方减少肺泡中白细胞的聚集,升高IL-10和TGF-β1,抑制IL-5和MUC5AC的产生,抑制气道黏液高分泌和炎症有关。
Objective To investigate the effect of Hua-Tan-Huo-Xue-Fang (HTHXF) on the number of leukocytes and the number of leukocytes in bronchoalveolar lavage fluid (BALF) induced by ovalbumin (OVA) IL-10, TGF-β1, MUC5AC expression and explore its possible mechanism of action in the treatment of asthma. Methods The sensitization solution containing OVA and aluminum hydroxide was injected intraperitoneally. The asthma model was induced by inhalation of OVA. On the 9th day of experiment, intragastric administration was given. The time of intragastric administration during the period of aerosolization was controlled at 1 hour before aerosolization. The mice were sacrificed on the 28th day and the number of leukocytes and the levels of IL-5, IL-10, TGF-β1 and MUC5AC in BALF were measured. Results Huatan Huoxue Decoction significantly reduced the total number of lymphocytes, neutrophils, eosinophils, monocytes and leukocytes in BALF, decreased the levels of IL-5 and MUC5AC, and increased the levels of IL-10 and TGF-β1. Conclusion The mechanism of Huatan Huoxue Recipe in treating asthma may be related to reducing the aggregation of leukocytes in the alveoli, increasing the levels of IL-10 and TGF-β1, inhibiting the production of IL-5 and MUC5AC and inhibiting the secretion and inflammation of airway mucus related.
