目的:观察中药柴贝止痫汤对海人酸(KA)致痫大鼠模型脑组织耐药蛋白MRP2(MRP2)的作用。方法:采用侧脑室注射KA的方法建立癫痫大鼠模型,分为正常组、假手术组、模型组、卡马西平组、中药组和中西药组共6组,分别于给药1个月及3个月后取新鲜脑组织进行蛋白质印迹法(Western Blot)以及实时荧光定量PCR(RT-PCR)来观察各组大鼠脑内MRP2表达的情况并比较组间差异。结果:与模型组比较,1月及3月正常组、假手术组大鼠海马组织中MRP2的表达显著降低(P<0.05),中药组及中西药组表达显著降低(P<0.05)。结论:长期服用卡马西平使MRP2在KA致痫大鼠脑组织中表达升高,而中药柴贝止痫汤可降低KA致痫大鼠脑组织中MRP2的表达。
Objective: To observe the effect of Chaibeixishi epilepsy decoction on MRP2 (MRP2) in the rat model of epilepsy induced by kainate (KA). Methods: A rat model of epilepsy was established by intracerebroventricular (icv) injection of KA. The model was divided into 6 groups: normal group, sham operation group, model group, carbamazepine group, traditional Chinese medicine group and western medicine group. Three months later, fresh brain tissue was taken for Western Blot and Real-time Fluorescent Quantitative PCR (RT-PCR) to observe the expression of MRP2 in rat brain and to compare the difference between groups. Results: Compared with the model group, the expression of MRP2 in the hippocampus of normal and sham-operated rats decreased significantly in January and March (P <0.05), and decreased significantly in the traditional Chinese medicine group and the Western medicine group (P <0.05). Conclusion: Carbamazepine prolonged MRP2 expression in the brain tissue of KA-induced epilepsy rats, while the Chinese medicine Chai-Bei Zhi epilepsy decoction can reduce the expression of MRP2 in the brain tissue of KA-induced epilepsy rats.
