目的:研究肝癌细胞核蛋白调控白蛋白"持家基因"转录调控序列对目的基因转录的作用。方法:分别从小鼠肝癌细胞、肝细胞和黑色素瘤细胞中提取核蛋白组分,用于体外启动白蛋白增强子/启动子的转录作用。结果:发现某些肝癌细胞和肝细胞核蛋白组分具有明显的转录作用,而细胞浆蛋白和其他肿瘤核蛋白组分没有转录作用,将同一肝癌细胞某些核蛋白组分合并可使转录活性增强。结论:本研究在基因转录调控水平部分解释了应用白蛋白增强子/启动子调控目的基因于肝癌细胞中特异表达的机理。
OBJECTIVE: To study the role of the transcriptional regulatory sequence of albumin “housekeeping gene” regulating transcription of target genes in liver cancer cell nuclear proteins. Methods: Nucleoprotein fractions were extracted from mouse hepatoma cells, hepatocytes and melanoma cells, and used to initiate the transcription of albumin enhancer/promoter in vitro. RESULTS: It was found that some hepatoma cells and hepatocyte nuclear protein components have obvious transcriptional effects, while cytoplasmic proteins and other tumor nuclear protein components have no transcriptional effect. Combining certain nuclear protein components of the same hepatoma cell can increase transcriptional activity. . Conclusion: This study partially explained the use of albumin enhancer/promoter to regulate the specific expression of the target gene in hepatoma cells at the level of gene transcription regulation.
