本文采用荧光分光光度法测定喜树碱在血及各组织中药物浓度,对喜树碱精氨酸盐静脉乳剂在小鼠体内的药代动力学及组织分布进行了研究。结果表明该制剂血药浓度经时过程为三室模型,其α、β、γ相半衰期分别为0.036h、0.487h、146.7h、Auc。→24为323.7μgh/ml,Cl为1.235ml/h。药物在肝组织的浓度明显高于其它组织,从而可以减少喜树碱用量而达到降低毒性提高治疗指数之目的。
In this paper, fluorescence spectrophotometry was used to determine the concentration of camptothecin in blood and tissues. The pharmacokinetics and tissue distribution of camptothecin arginine salt emulsion in mice were studied. The results showed that the preparation of the plasma concentration over time as a three-compartment model, the α, β, γ phase half-life were 0.036h, 0.487h, 146.7h, Auc. → 24 is 323.7 μgh / ml, Cl is 1.235 ml / h. The concentration of the drug in the liver tissue is obviously higher than other tissues, so that the amount of camptothecin can be reduced to reduce the toxicity and improve the therapeutic index.
