目的探讨胎盘生长因子(PlGF)在脑膜瘤中的作用机制,以进一步明确脑膜瘤预后影响因素,指导临床治疗,提高患者生存率。方法本研究采用免疫组化和原位杂交检测方法对105例不同类型脑膜瘤的病理组织、免疫表型及基因水平表达特点进行系统地观察、分析。结果免疫组化结果显示PlGF在各型脑膜瘤中均有表达,其在间变型脑膜瘤中的表达明显高于其他两型(P<0.05)。相关分析发现,在间变型脑膜瘤中PlGF表达与MVD值呈明显正相关(r=0.392,P<0.01);且VEGF与PlGF亦存在正相关(r=0.486,P<0.05)。原位杂交检测结果,PlGFmRNA杂交反应产物在三型脑膜瘤中均存在,但其三者之间并无明显差异。结论PlGF在三型不同性质脑膜瘤中均有不同程度的表达;VEGF与PlGF在间变型脑膜瘤血管生成中发挥协同作用,对于今后进一步应用抗血管新生药物治疗脑膜瘤、提高患者生存率提供了一条新途径。
Objective To investigate the mechanism of placental growth factor (PlGF) in meningioma, to further clarify the influencing factors of prognosis of meningioma, to guide the clinical treatment and to improve the survival rate of patients. Methods The immunohistochemical and in situ hybridization methods were used to systematically observe and analyze the pathological features, immunophenotype and gene expression in 105 cases of different types of meningioma. Results The results of immunohistochemistry showed that PlGF was expressed in all kinds of meningioma and its expression was significantly higher in meningioma than in other two types (P <0.05). Correlation analysis showed that there was a positive correlation between the expression of PlGF and MVD (p = 0.392, P <0.01), and the positive correlation between VEGF and PlGF (r = 0.486, P <0.05). In situ hybridization results, PlGFmRNA hybridization products in three types of meningiomas are present, but there was no significant difference between the three. Conclusions PlGF is expressed in different degree in different types of meningiomas of different types. VEGF and PlGF play a synergistic role in angiogenesis of anaplastic meningioma, which provides a new way to further apply antiangiogenic drugs to meningioma and improve the survival rate of patients A new way.
