抗血管紧张素Ⅱ受体1型自身抗体致大鼠胸主动脉收缩的作用

目的:分析抗血管紧张素Ⅱ受体1型自身抗体(AT1-AA)对子痫前期患者血压的影响。方法:利用MAb Trap试剂盒对子痫前期患者抗体阳性者血清中的抗体IgG进行提取和纯化,用血管环技术检测纯化后的抗体对大鼠胸主动脉收缩功能的影响。结果:AT1-AA剂量依赖性地引起大鼠胸主动脉的收缩,0.01μmol/L、0.1μmol/L和1.0μmol/L的AT1-AA可使大鼠胸主动脉血管环的最大收缩张力从对照组的1.02±0.04 g分别增加到1.06±0.05 g、1.28±0.09 g和1.72±0.17 g。结论:AT1-AA对大血管具有直接的收缩效应,可能是引起子痫前期的原因之一。 Objective: To analyze the effect of anti-angiotensin II receptor type 1 autoantibody (AT1-AA) on blood pressure in preeclampsia patients. Methods: Antibodies IgG in serum of patients with preeclampsia were extracted and purified by MAb Trap kit. The effect of purified antibody on the contractile function of thoracic aorta of rats was detected by vascularization. Results: AT1-AA induced a dose-dependent contraction of the thoracic aorta in rats. AT1-AA at concentrations of 0.01μmol / L, 0.1μmol / L and 1.0μmol / L significantly decreased the maximum systolic tension of the thoracic aortic rings 1.02 ± 0.04 g in the control group increased to 1.06 ± 0.05 g, 1.28 ± 0.09 g and 1.72 ± 0.17 g, respectively. CONCLUSION: AT1-AA has a direct systolic effect on the large blood vessels and may be one of the causes of preeclampsia.
